ABSTRACT

OBJECTIVE To investigate the underlyingmechanism on the association of red blood cell and gut microbiota in rats induced by High-Fat Diet(HFD).METHODS A total of 36 male Sprague-Dawley rats (180±20g) were randomly divided into two groups. The control group (n=10) was given a normal chow diet(10% calories of fat),and the High-fat diet group(n=26)was given a HFD(60% calo-ries of fat).We recorded body weight,length and detected serum glucose,serum lipids and insulin ev-ery two weeks.The fresh arterial blood was collected during the experiments and blood gases were measured immediately (Radiometer Medical ApS, Denmark).Thehematocrit (Hct) and partial pressure of oxygen(pO2)were detected by the sensor cassette,following themanufacturer′s instructions.The de-tection method was conductivity measurements and current method, respectively. The feces from ce-cum were analyzed by 16S rRNA gene high-throughput sequencing(Illumina Miseq,USA). RESULTS According to the insulin resistance(IR),body weight and body length,the model group was divided into two small groups.(1)IR group,in which IR,body weight and body length were higher than the control group (P<0.05). (2) un-IR group, body weight and body length were higher than the control group (P<0.05),but the IR was not significantly different.In addition,the levels of hematocrit(Hct),checktotalhe-moglobin (ctHb) and check total blood oxygen content (ctO2) showed significantly increased in the IR group when compared with the control group (P<0.05), however, the pO2was not statistically signifi-cant. Furthermore, we identified that the genus Lactobacillus was moderate positive correlation with Hct,ctHb and ctO2(P<0.05).Compared with the control group,the relative abundance of the Lactoba-cillus was significantly lower in IR group(P<0.05).CONCLUSION The high-fat diet induced rats′local tissue hypoxia under the red blood cell increasing,oxygen partial pressure constant and the reduction of Lactobacillus′abundance might be caused by aerobic oxidation and glycolysis inhibition in the meantime.

ABSTRACT

OBJECTIVE To explorethe correlation betweenhypoxiaand insulin resistance bythe blood gas index in high-fat diets-induced obese rat model. METHODS 36% of high-fat diets were fed to SD male rats for 12 weeks. The model group was divided into IR group and non-IR group with the HOMA-IR index of the 12th week,and the abdominal aorta blood was taken for blood gas analysis. RESULTS The HOMA-IR index,Hct,ctHb and ctO2in IR group were significantly higher than those in normal group andnon-IR group(P>0.05),simultaneously no significant difference in pO2,pCO2and sO2 between tree groups.CONCLUSION Circulating blood of obese rat with insulin resistance is normoxia,accompanied by higher Hct,tHb and ctO2,which may be due to the higher blood viscositand the self-regulation of chronic hypoxia in the body.

ABSTRACT

Aim To investigate the effect of petroleum ether extract of Eclipta prostrata(PEEEP) on biochem-ical parameters such as blood glucose, insulin, total glycerides,total cholesterol,creatinine,urea nitrogen, uric acid levels and histopathology of kidney in STZ in-duced diabetic rats. Methods The model of type 2 diabetic rats was established by feeding with high ener-gy diet for 8 weeks and injecting streptozotocin(30 mg ·kg-1), after making the model successfully, the rats were randomly divided into model control group, metformin positive control group and experimental group, and gavaged with distilled water, metformin (400 mg·kg-1) and different dose of PEEEP (100, 200,400 mg·kg-1) for 4 weeks, respectively. The contents of blood glucose,insulin,total glycerides,to-tal cholesterol, creatinine, urea nitrogen, uric acid were detected by corresponding kit, the pathological changes of kidney were observed by light microscopy, and the expression of TNF-α protein was detected by Western blot. Results Compared with model control group, PEEEP could reduce blood glucose, total glyc-erides, total cholesterol, creatinine, urea nitrogen, u-ric acid and increase the content of insulin in a dose-dependent manner (P <0.01 or P <0.05). It also could alleviate the pathological damage of kidney tis-sues and down-regulate the expression of TNF-α in kidney tissues. Conclusions PEEEP could effectively improve related biochemical parameters in streptozoto-cin induced diabetic rats, and it has protective effect on the kidney, providing the theoretical basis for fur-ther exploitation and utilization of Eclipta prostrata.

ABSTRACT

Objectives:To investigate the clinical efficacy and influencing factors on clinical outcome of extracorporeal membrane oxygenation (ECMO) combined with percutaneous coronary intervention (PCI) on cardiac arrest patients due to acute myocardial infarction (AMI). Methods:The clinical data of 37 cardiac arrest patients post AMI who underwent ECMO combined with emergency PCI therapy strategies from January 2015 to January 2017 were retrospectively analyzed. The clinical data of the surviving group (17 cases) and the death group (20 cases) were compared and the outcome determinants were investigated. Results:37 patients were successfully treated with emergency PCI with the help of ECMO device, and the successful operation rate was 100%. The survival rate was 45.9% (17/37). There was no significant difference in gender, age and past history between the two groups (P>0.05). The proportion of right coronary artery lesion was the highest (47.06%) in the surviving group, and the percent of the left anterior descending coronary artery lesion was the highest (60.00%) in the death group. Most patients (70.0%) in the death group had three vessel lesions and around half the patients (52.9%) in the surviving group had two-vessel lesions. The time of traditional cardiopulmonary resuscitation ([31.4±5.2] min vs [54.6±25.7] min),the time from cardiac arrest to ECMO ([47.5±19.5] h vs [93.6±60.5] h) were significantly shorter, while CCU time (16.0[8.7, 32.6] d vs 4.0[2.0, 12.0] d) was significantly longer in the surviving group compared to the death group (all P<0.05). The mean arterial pressure at 24 h and 48 h after ECMO was significantly higher in the surviving group (89.6±21.9,87.9±19.4) than in the death group (71.8±19.3, 63.7±18.6) (both P<0.05). Unconditional logistic regression analysis showed that left anterior descending artery lesion (OR=0.723, 95%CI:0.516~0.947), higher lesion vessel number (OR=1.638,95%CI:1.107~1.729), longer cardiopulmonary resuscitation time (OR=0.712, 95%CI:0.436~0.973), prolonged cardiac arrest to ECMO placement time (OR=0.698, 95%CI:0.411~0.859) were risk factors of death, and the higher MBP at 48 h after ECMO (OR=0.672,95%CI:0.326~0.693) was the protective factor of death (all P<0.05) in this patient cohort. Conclusions:ECMO combined with emergency PCI is safe and can improve the success rate of rescue in patients with cardiac arrest after acute myocardial infarction.

ABSTRACT

The function of the nervous system is largely determined by the communication between neurons and glial cells.The interaction of complex signaling pathways between neurons and glial cells involves in a variety of signaling molecules.Purines and the purinergic receptors play a crucial role in the process of the interaction.For a long time, the study on the central nervous system is mainly focused on neurons, while glial cells are mainly used as support cells or in response to brain injury to provide a guiding role.The latest research shows that glial cells play an important role in the occurrence and development of nervous system diseases.We will summarize the function of purinergic receptors in the interaction of neurons and glial cells and in neurological diseases.

ABSTRACT

@#AIM: To explore the efficacy of Ex-press glaucoma drainage device on open-angle glaucoma. <p>METHODS: We collected the clinical data of those patients who were diagnosed as open angle glaucoma and had undergone Ex-press glaucoma drainage device implantation or trabeculectomy in the Second Hospital of Anhui Medical University from January 2014 to May 2016. The data of patients was collected and used to make a comparison involving sex, ages, best corrected visual acuity(BCVA), intraocular pressure(IOP), the amount of corneal endothelium cells and complications before and after operations. <p>RESULTS: There was no significantly different(<i>P</i>>0.05)between the Ex-press group and trabeculectomy group on postoperative BCVA at 6mo. Both groups had obvious effects on the drop of IOP after operations(<i>P</i><0.001), but there was no difference between the two groups at any time after operations. Ex-press group had no significant difference at 6mo while it had in the trabeculectomy group about the amount of corneal endothelium cells comparing with preoperative. The incidence rate of overall complications about Ex-press group and trabeculectomy group was 15% and 43% respectively, it had a significant difference between two groups(<i>P</i><0.05). <p>CONCLUSION: The Ex-press glaucoma drainage device had similar efficacy with lower complication rate and smaller influence of corneal endothelium cells comparing with trabeculectomy.

ABSTRACT

The samples of Huangqi injection (HI) were analyzed by liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (LC-TOF-MS), and both positive and negative ion modes were employed to obtain the LC-TOF-MS analysis information of chemical compounds in HI. Then the mass defect filtering (MDF) approach, which was developed based on the previously published articles, was utilized to rapidly screen the astragalosides from the obtained LC-TOF-MS data. Each screened astragaloside was confirmed by the presence of no less than 2 quasi-molecular ions. All the screened astragalosides were then tentatively assigned according to the parent ion and daughter ion information. Finally, a total of 62 astragalosides were screened and characterized from the HI samples, including 15 new detected ones. The identification results indicated that acetylation, hydrogenation, dehydrogenation, methoxylation and hydration might be the major conversion reactions involved in the formation of the astragalosides. The LC-TOF-MS-based MDF approach was proved to be a feasible and efficient tool to screen the chemical constituents in complex matrices such as herbal medicines.

ABSTRACT

Aim To study the protective effect of the naringin on chemotactic factor CX3 CL1 of human umbilical vein endothelial cells ( HUVEC) induced by high glucose. Methods The effect of different concen-trations of naringin on HUVEC cell viability was deter-mined by MTS. HUVECs were divided into 4 groups:① control group, ② high glucose group, ③ naringin group and④high glucose treated with naringin group. After treatment for 5 days, the concentration of nitric oxide ( NO ) in the culture medium was measured by nitrate reductase; intracellular reactive oxygen species ( ROS) was analyzed with fluorescence probe; the ex-pressions of CX3 CL1 mRNA and protein were deter-mined by the reverse transcription PCR ( RT-PCR ) and Western blot ( WB ) . Results NO content in the culture medium of high glucose group was markedly de-creased, which could be increased by naringin. Com-pared with the control group, intracellular ROS in the high glucose group was drastically elevated, but narin-gin decreased the elevated ROS induced by high glu-cose. The results of RT-PCR and WB showed that nar-ingin could downregulate the increased expressions of CX3CL1 mRNA and protein induced by high glucose. Conclusion Naringin has protective effect on the in-jury of the HUVEC induced by high glucose, which is associated with reducing the expression of CX3 CL1 and the antioxidative and anti-inflammatory action.

ABSTRACT

A method was proposed for the separation and determination of paraffin waxes in food by HPLC-evaporative light scattering detection (ELSD). A normal-phase column was used to separate nonparaffinic and paraffinic materials without resolving the latter into individual components. The t-test method was adopted for the evaluation of mean difference between response factors of n-alkanes in paraffin waxes on ELSD detector. No mean difference was obtained between response factors, which can be used for quantitative determination of paraffin waxes in food. The determination results obtained by HPLC-ELSD were compared with those by GC-MS. The linear range for the determination of paraffin waxes was in the range from 10 to 500 mg/L with a correlation coefficient of 0.9988, and the limit of detection was 1.0 mg/L. With the spiking level of 10, 50 and 100 mg/kg, the recovery ranged from 84.6% to 105.4% and the relative standard deviation ranging from 5.4% to 7.2%. The proposed method is simple, fast and sensitive.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the safety and efficacy of drug-eluting stent (DES) implantation in selective patients with left main coronary artery disease.</p><p><b>METHOD</b>From October 2002 to November 2007, 44 consecutive patients underwent percutaneous coronary interventions (PCI) on left main coronary artery lesions, including 5 patients with concurrent left ventricular dysfunction (ejection fraction<40%), 2 with chronic respiratory dysfunction and 5 with chronic renal failure. The findings in coronary angiography, procedural success rate, severe complications and the follow-up results of the patients were analyzed.</p><p><b>RESULTS</b>The immediate procedural success rate was 100% in these patients without any severe complications. No non-fatal acute myocardial infarction or emergency coronary artery bypass grafting (CABG) was performed and death occurred in none of the cases during hospitalization. In the follow-up period for 14.2-/+9.3 (6-65) months after PCI, no subacute or late thromboses were found. One patient died from heart failure 4 months after PCI, and 6 patients (13.6%) experienced recurrent angina. Thirty-seven patients (84.1%) were free of any major cardiovascular events (MACE) after the procedure. A repeat coronary angiography was performed in 35 patients (79.5%) within 6 months after PCI, and 3 (8.6%) of them were confirmed to have restenosis, including 1 patient with distal bifurcation restenosis who were subsequently treated with CABG and two patients with side-branch ostium restenosis managed with cutting balloon dilation.</p><p><b>CONCLUSIONS</b>Implantation of drug-eluting stents is safe and effective for management of left main coronary artery disease with good immediate and long-term outcomes.</p>
